Chemical Installation and Endogenous Cycling in Protein S-Palmitoylation: Interpreting Gain-of-Function Evidence

Abstract

Site-specific chemical installation of a fatty-acyl group offers a direct way to ask what a modified protein can do. It also changes the interpretation problem: a chemically installed mimic need not reproduce the dynamics, occupancy, or molecular interactions of a reversible endogenous modification. This conceptual analysis examines live-cell gain-of-function evidence through the chemical fatty-acylation study of Li and colleagues. It separates successful installation, membrane association, downstream signaling, and correspondence with native S-palmitoylation. The proposed validation strategy combines site controls, chemical controls, temporal observations, and independent biological replication. A bounded role for efficient adaptation concerns extraction and organization of experimental records, not replacement of a biochemical assay. No proteins were modified and no live-cell experiment or molecular model was run here. The article develops an evidence framework for deciding which functional claims a site-specific perturbation can support.
